藏羚羊成纤维细胞与受体卵母细胞的化学辅助融合方法

本发明涉及一种藏羚羊成纤维细胞与受体卵母细胞的化学辅助融合方法,该方法包括以下步骤:(1)供体细胞和受体细胞的选择:供体细胞选择藏羚羊成纤维细胞,并将其培养在24孔板中;受体卵母细胞选择山羊、牦牛、黄牛或奶牛卵母细胞中的任意一种;(2)制作体积为25μL的融合微滴;(3)供体细胞与受体卵母细胞的融合:依次将受体卵母细胞移至T20、植物血凝素、T2B、融合液微滴内进行操作,形成两个卵母细胞夹杂一个供体细胞的复合体;将复合体移至融合液内平衡,再至融合槽内进行电融合即可。本发明有效地提高了细胞融合效率、降低了操作难度、缩减了操作时间,从而优化了手导式克隆技术细胞融合的技术环节。 | The invention relates to a chemical auxiliary fusion method of Tibetan antelope fibrocyte and receptor ovarian ovum. The method comprises the following steps of: (1) selecting a donor cell and a receptor cell, wherein the donor cell is selected from the Tibetan antelope fibrocyte and cultivated in a 24-pore plate, the receptor ovarian ovum is selected from any one of goat, yak, cattle or dairy cow oocyte; (2) manufacturing fused droplet with the volume of 25uL; and (3) fusing the donor cell and the receptor cell: transferring the receptor oocyte to T20, plant hemagglutinin, T2B and fused droplet in sequence for operation to form a compound with two oocytes and one donor cell, transferring the compound to the fusion liquid for balance and then to a fusion tank for performing electric fusion. The invention effectively improves the cell fusion efficiency, reduces the operation difficulty, shortens the operation time and optimizes the technical link of cell fusion by the hand brake-type clone technology.
