This strain carries the "RIP-Cre" transgene construct (containing a 668 bp fragment of the rat insulin II promoter, Cre recombinase with a nuclear localization sequence, and a 2.1 kb fragment from the human growth hormone gene). Hemizygous mice carrying this transgene are phenotypically normal and overexpresss cre specifically in pancreatic beta cells. This transgene strain is used in combination with mice carrying floxed targeted mutations to create various pancreatic beta cell-specific gene knockouts using the "Cre-lox" system. Results from several different laboratories have shown that this transgenic strain is at least 85% efficient in achieving pancreatic beta cell-specific recombination. It should also be noted that the transgene in this line has been found to be expressed at a low level in the hypothalamus. In some cases this has resulted in a phenotype due to deletion of the floxed allele in this region of the brain. It has also been shown that these transgenic mice may spontaneously develop glucose intolerance and impaired insulin secretion developing at 6-8 weeks of age. It is recommended that users include naive "RIP-Cre" mice (i.e., those not bred to a floxed mutant) among the controls used in experiments.(来自Jackson网站)

→ A targeting vector containing Cre recombinase, the rabbit beta-globin intron /poly A signal sequence, and an FRT-flanked neomycin resistance gene, was used to disrupt exon 2 of the Cd19 gene and to express cre under the regulation of the endogenous promoter. Herpes simplex virus thymidine kinase gene was placed 3' of the Cd19 sequence to allow for the selection against random integration. The construct was transfected into 129P2/OlaHsd-derived E14-1 embryonic stem cells. Correctly targeted ES cells were injected into 129/Sv blastocysts. Homozygous mutant mice are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. Homozygotes have a deficiency in the B-1 subset of B-lymphocytes along with a concomitant reduction in serum IgM. Their ability to respond to T-cell-dependent antigens is severely impaired, and they fail to form splenic germinal centers.
In addition to disrupting the targeted gene, the targeting construct also introduced acre cassette into exon 2 of the targeted gene, effectively placingcre expression under the control of the endogenous promoter. The Cd19 promoter specifically directs cre expression at the earliest stages and throughout B-lymphocyte development and differentiation. Although homozygous mutant mice are Cd19-deficient, heterozygous mice are phenotypically normal, and can be used for specific deletion of loxP-flanked (floxed) targets in B-lymphocytes.
In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. This transgenic strain expresses Cre under the control of the Lck (lymphocyte protein tyrosine kinase) promoter, enabling thymocyte-specific excision of loxP-flanked sequences of interest. Homozygous mice are viable and have no major defects.
When crossed with floxed Galnt13 mice (see Stock No. 006896), excision occurs at high efficiency in thymocytes. Splenic CD4 T cells show excision at somewhat lower rates (66-83%).
The Lck gene is primarily expressed by T lymphocytes where it phosphorylates tyrosine residues of proteins involved with intracellular signaling pathways. Associating with CD4 and CD8, LCK is part of a cascade of responses to antigen that control the proliferation and activation of T cells. It plays a key role in the selection and maturation of developing T-cells in the thymus and in the activity of mature T-cells.(来自Jackson网站)

→ In 1966, outbred Swiss mice at the National Institutes of Health were selectively bred for either resistance or sensitivity to histamine challenge post pertussis vaccination. At the eighth generation of inbreeding (in the early 1970s), the sensitive line, HSFS/N, was found to carry the Fv1b allele which confers sensitivity to the Friend leukemia virus B strain. The FVB/N strain resulted from inbreeding this line for the Fv1b allele. In 1988 FVB/N mice were imported from NIH to Dr. Taketo at The Jackson Laboratory and in 1991 these were re-derived at F50 into the foundation stocks facility at The Jackson Laboratory. FVB/NJ was inbred for the Fv1b allele which confers sensitivity to the Friend leukemia virus B strain. Due to the prominent pronuclei in their fertilized eggs and the large litter size, FVB/NJ mice are commonly used for transgenic injection. Compared to many other inbred strains, FVB/NJ is highly susceptible to asthma-like airway responsiveness with significant generation of antigen-specific IgE. Despite having the H2q MHC haplotype, FVB/NJ are resistant to collagen-induced arthritis. This resistance stems from coding polymorphisms in Tcra-V11.1 and a genomic deletion of some Tcrb-V genes that includes Tcrb-V8.2. FVB/NJ have higher than average activity, anxiety, and basal body temperature, low stress-induced hyperthermia, and are homozygous for the Pde6brd1 allele, which results in early onset retinal degeneration. Although FVB/N typically do not develop spontaneous tumors, they are highly susceptible to chemically induced squamous cell carcinomas with a high rate of malignant conversion from papilloma to carcinoma.(来自Jackson网站)

免疫:在普通饲养条件下三月龄鼠血清免疫球蛋白量为1000ug/ml左右,仅相当C57BL/6,C3H/He和BALB/c的1/2。其中,IgM值较高,而IgG为低值。在IgG各亚类中,IgG1高低,IgG2最低。肿瘤:对DBA/1 的大部分移植瘤有抗性。雌鼠白血病发病率为34%,雄鼠为18%,经产母鼠乳腺癌发生率为50- 60%,雌雄鼠中均有淋巴瘤生长。微生物和寄生虫:对疟原虫、利什曼原虫有抗力。对猫后睾吸虫、曼氏血吸虫较敏感。对白色念球菌有抗力,由于具有Hc0等位基因, 对新型隐球菌有抗力。生理:红细胞多。血压较低。维生素K缺乏,氯仿和氧化乙烯引起的死亡率高。肾上腺脂质贮存少,心脏有钙盐沉着。具低嗜酒性及吗啡嗜好。对百日咳组织胺易感因子敏感。病理:听源性癫痫发作率在35日龄时为100%,55日龄时为5%,约一半动物肝可出现由巨噬细胞构成的蜡样质的肉芽肿。

Mice homozygous for the obese spontaneous mutation, (Lepob; commonly referred to as ob or ob/ob), are first recognizable at about four weeks of age. Homozygous mutant mice gain weight rapidly and may reach three times the normal weight of wild-type controls. In addition to obesity, mutant mice exhibit hyperphagia, a diabetes-like syndrome of hyperglycemia, glucose intolerance, elevated plasma insulin, subfertility, impaired wound healing, and an increase in hormone production from both pituitary and adrenal glands. They are also hypometabolic and hypothermic. The obesity is characterized by an increase in both adipocyte number and size. Adipose tissue transplants in Lepob homozygotes protect them from obesity, normalize insulin sensitivity, and restore fertility. Although hyperphagia contributes to the obesity, homozygotes gain excess weight and deposit excess fat even when restricted to a diet sufficient for normal weight maintenance in lean mice. Hyperinsulinemia does not develop until after the increase in body weight, and probably results from it. Homozygotes have an abnormally low threshold for stimulation of pancreatic islet insulin secretion even in very young pre-obese animals. Female homozygotes exhibit decreased uterine and ovarian weights, decreased ovarian hormone production and hypercytolipidemia in follicular granulosa and endometrial epithelial tissue layers (Garris et al., 2004).
As is the case with mice carrying the diabetes mutation (Leprdb), manifestation of the diabetic syndrome is strikingly dependent on genetic background. Hyperglycemia is only transient, (subsiding around 14 to 16 weeks) on the C57BL/6J background. On the C57BLKS background, obese homozygotes (Stock No. 000696) become severely diabetic with regression of islets and early death. Injection of recombinant leptin into obese homozygotes sharply reduces body weight, decreases food intake, increases energy expenditure, and restores fertility in male mice.
Leptin can regulate bone mass through a central, neuroendocrine signaling pathway. Similar to the effects of aging in humans, homozygotes exhibit muscle hypoplasia (quadriceps), increased marrow adipogenesis and decreased bone mass in the hindlimbs (Hamrick et al., 2004) (来自Jackson网站)

Mice homozygous for the Igf1tm2Ts mutation (also referred to as the Igf1m/m midi targeted insertion) are viable and fertile, but are small in size, reaching only approximately 60 - 65% the weight of their wild type siblings. Serum levels of IGF-1 in these mutants is 30% of wild type values. They have no gross histological abnormalities. Arterial blood pressure is higher in Igf1m/m mice than in wild type controls. The Donating Investigator reports that homozygotes are subfertile but viable, with approximately 50% of homozygous animals dying before weaning.(来自Jackson网站)
